The effects of concurrent administration of Diphenyl-dimethyl-dicarboxylate with Diltiazem or Propranolol on CCl_4-induced liver injury


Abstract
The effects of concurrent administration of Diphenyl-dimethyl-dicarboxylate(PMC) with Diltiazem (a calcium channel blocker) or Propranolol (¥â-adrenergic antagonist) on CCI_4-induced liver injury were determined.
A significant decrease in both transaminase(s-GOT, s-GPT) and malondialdehyde level was found. Futhermore, on histopathologic examination, and important decrease was observed in severe necrosis and infiltration of inflammatory cells around the central vein. Ballooning degeneration of hepatocytes were markedly improved.
These results show the following two additive actions. First action is free radical scavenging action of PMC on trichloromethyl peroxy free radical which are reactive intermediates of CCI_4, and second one is inhibitory action of Diltiazem or Propranolol against the entry of extracellular Ca^2+ into hepatocytes which is caused by the partial reactive intermediates of CCI_4 left over the free radical scavenging action of PMC.
